Salvarsan – the First Chemotherapeutic Compound

چکیده
Early History If defined as the use of a specific compound discovered as the result of a systematic search for a cure for a specific disease, then chemotherapy is less than a century old. The first example was the introduction in 1910 by Paul Ehrlich of an organoarsenic compound he named Salvarsan, which provided the first real cure for the extremely unpleasant disease syphilis (caused by the parasitic spirochete Treponema pallidum). As is well-known, the compound was also called Ehrlich 606 because it was the 606 compound tested by Ehrlich in conjunction with his colleagues Bertheim (synthesis) and Hata (biological testing) (Figs. 1 & 2).

The composition of Ehrlich's salvarsan: resolution of a century-old debate.
In 1910, Ehrlich introduced the compound 3-amino-4hydroxyphenylarsenic(i) (salvarsan, arsphenamine, or Ehrlich 606) as a remedy for syphilis, a disease caused by the spirochaete bacterium Treponema pallidum. His methodical search for a specific curative for an identified disease can be regarded as the introduction of targeted chemotherapy. Despite its long history and importance, the actual str...

Although arsphenamine’ has within recent years attained the leading place in the treatment of syphilis, its condensation product with sodium formaldehyde sulfoxylate, known as neoarsphenamine, is beginning to find preference with physicians. There are several reasons why it is considered superior. First, it is easier to prepare a solution of neoarsphenamine for intravenous injection, since it i...

The researches of Ehrlich and his coworkers during the past decade, which led to the discovery of salvarsan, have created a new procedure in the search for remedies for the control of infectious diseases. The discovery of this drug was the result of a logically conducted development of the chemistry of organic arsenic compounds in the directions indicated by their biological properties. This ne...
متن کاملRational Approaches to Antibacterial Discovery: Pre-Genomic Directed and Phenotypic Screening
Early antibacterial discovery might be divided into the synthetic and antibiotic traditions. The salvarsan-prontosil strand led to exploration of antimetabolite and other chemotherapeutic approaches, while penicillin’s discovery led to the fruitful exploitation of natural products produced by microorganisms. With the investigation of the mechanisms of action of synthetic and antibiotic agents a...

bacteria (and not by host cells), it could be the ‘magic bullet’ of his dreams7. After testing over nine hundred dyes and chemical compounds, the 606 compound (latter named Salvarsan) was found to be effective against the etiological agent of syphilis; but unfortunately such compound toxicity against the host was evidenced. While the ‘606’ comprised a huge advance in the war on microbes, it bec...
